Parallel setup of advanced P-SUBmerged™ with Clotho Tower
For applications involving mammalian cell retention for expressing mAbs - low RPM and preferably no bursting aeration bubbles is mandatory. The cell lines are fragile and in a perfusion setup benefit from.
- No HFF fouling – freedom to select broth velocities flushing out debris from inside the lumen regularly
- No dead volumes – skip all the hoses and plastic connectors, SUBmerged and integrate the HFF 100 % with Thalia diaphragm pump
- No local cell death - inverted Thalia pump above the HFF and SUB cannot accumulate cells
- Reduced shear force – remove all hoses and fittings and SUBmerge the HFF directly into the media inside the SUB
We introduce the SUBmerged™ - the customized SepraPor HFF inlet are arranged SUBmerged in the broth inside the SUB. The HFF in the opposite end directly coupled to the Thalia™ diaphragm pump with no coupling, hoses, etc. The Thalia diaphragm Single-Use-Pump (SUP) is LASER guided and its unique performance is programmed selectively into the Clotho™ drive unit. The "CellRetention-300-DASbox" perfusion bioreactor available from sister company PerfuseCell™.
The above example is a CellRetention HFF SUBmerged designed specifically for the DASbox. The OD76 mm BioBLU® is heavily customized and equipped with the SepraPor® HFF and the Thalia™ SUP. DASbox takes care of agitation, aeration, pH, DO and media supply. The Clotho™ Tower controls Thalia which takes care of ATF harvest and the HFF cleaning process and hereby protects the HFF. The pressurized air and vacuum required for Clotho is delivered by dual Alagonia and one Sarpedon.
For the Modular PCS we offer a kit converting the 250 ml batch SUB to become P-SUBmerged. The Thalia diaphragm Single-Use-Pump is LASER guided and its unique performance is programmed selectively into the Clotho drive unit. The "CellRetention-250-kit-Modular" perfusion bioreactor available from sister company PerfuseCell™.
Any end-user can in their hood install the kit and easily go from SUB to P-SUB. The Modular PCS takes care of agitation, aeration, pH, DO and media supply. The Clotho Tower controls Thalia which takes care of ATF harvest and the HFF cleaning process and hereby protects the HFF. The pressurized air and vacuum required for Clotho is delivered by dual Alagonia and one Sarpedon - part of the Clotho Tower.
